扩增产物于0.8%的琼脂糖凝胶中电泳,在BioSenSC805型全自动凝胶成像系统的Analysis工具栏中,点击消除背景图标以消除图像本底的干扰;点击自动检测所有泳道的条带图标以进行各泳道内条带检测;点击相对分子质量计算图标,以Marker泳道内的Marker条带的相对分子质量为标准设定相对分子量曲线图,关闭Marker窗口,则会显示各个条带的相对分子质量,统计各泳道内的条带数。
2 结果与分析

2.2 亲本甜荞RAPD标记的多样性分析
根据筛选出的5个随机引物的RAPD扩增结果,可以对每对杂交组合中的两个亲本进行扩增条带的多样性分析,两对杂交组合亲本中各引物的扩增情况与相似度系数见表3和表4。
3 讨论
此外,本研究以重组自交系亲本为研究对象进行RAPD分析,发现自交系群体亲本之间存在较多的特异RAPD谱带,也可为以重组自交系为研究对象进行的相关农艺学特征遗传及分子标记研究奠定基础。
